mKATP activity, and used this assay probe several aspects of mKATP function. The following key observations were made. (1) Time-dependent run down of mKATP activity was reversed by phosphatidylinositol-4,5bisphosphate (PIP2). (2) Dose responses of mKATP to nucleotides revealed a UDP EC50 of 20 mol/L and an ATP IC50 of 5 mol/L. (3) The antidepressant fluoxetine (Prozac) inhibited mKATP (IC50 2.4 mol/L). Fluoxetine also blocked cardioprotection triggered by IPC, but did not block protection triggered by a

Neuroprotective Effect of Mitochondrial Katp Channel Opener Upon Neuronal Cortical Brain of Rat Population

Purpose: So far there is no effective drug therapy to prevent neuronal loss after brain stroke. In the present study we studied effects of The Mitochondrial K-ATP channel regulators on neuronal cell population and neurological function after ischemia reperfusion in the rat. Materials and Methods: Rats temporarily subjected to four vessels occlusion for 15 minutes followed by 24 hours reperfusi...

Biophysical and electropharmacological properties of single mitoKATP channel in rat brain mitochondrial inner membrane

Introduction: Different ATP-sensitive potassium channels have been detected in the mitochondrial inner membrane of cells. They are suggested to be involved in cell processes including cell protection. Here, we characterized the biophysical and electropharmacological properties of a KATP channel in the brain mitochondrial inner membranes. Methods: After removing and homogenizing the rat brain...

Mitochondrial ATP-dependent potassium channels: novel effectors of cardioprotection?

BACKGROUND Brief interruptions of coronary blood flow paradoxically protect the heart from subsequent prolonged ischemia. The basis of such endogenous cardioprotection, known as "ischemic preconditioning," remains uncertain. Pharmacological evidence has implicated ATP-dependent potassium (KATP) channels in the mechanism of preconditioning; however, the effects of sarcolemmal KATP channels on ex...

Evidences for a new cation channel in the brain mitochondrial inner membrane

Introduction: Previous studies and our works have indicated several cation channels in the rat brain mitochondrial inner membrane. In this work, we report the single-channel characterization of a cation channel from the rat brain mitochondrial inner membrane incorporated into a planar lipid bilayer. Methods: After removing and homogenizing the adult rat brain, its supernatant was centrifuged...

The impact of ATP-sensitive K+ channel subtype selectivity of insulin secretagogues for the coronary vasculature and the myocardium.

Insulin secretagogues (sulfonylureas and glinides) increase insulin secretion by closing the ATP-sensitive K+ channel (KATP channel) in the pancreatic beta-cell membrane. KATP channels subserve important functions also in the heart. First, KATP channels in coronary myocytes contribute to the control of coronary blood flow at rest and in hypoxia. Second, KATP channels in the sarcolemma of cardio...
